چکیده | ||
سیانوباکتریها بهعلت تولید انواع متابولیتهای ثانویه و ترکیبات ضدمیکروبی موردتوجه بسیاری از محققان قرارگرفتهاند. این تحقیق بهمنظور بهینهسازی محیط کشت BG11 با هدف افزایش تأثیر ضدقارچی سیانوباکتری Cyanobacteriumsp.PGU1 علیه دو قارچ بیماریزای گیاهی Fusarium oxysporum f. sp. lycopersici و Rhizoctonia solani AG2-2-IIIB انجام شد. سیانوباکتری جداسازیشده از آب خلیجفارس با دو روش ریختشناختی (مورفولوژیک) و تعیین توالی 16S rRNA با عنوان Cyanobacterium sp. PGU1 شناسایی شد. بهینهسازی محیط کشتBG11، با تغییر میزان منابع نیتروژن و فسفات موجود در محیط کشت، هرکدام در سه سطح، شامل 75/0، 5/1 و 3 گرم در لیتر برای NaNO3 و میزان 02/0، 04/0 و 08/0 گرم در لیتر برای K2HPO4انجام شد و تأثیر این مقادیر بر فعالیت ضد قارچی سیانوباکتری بررسی شد. تأثیر ضدقارچی عصارۀ متانولی بهدستآمده از کشت خالص سیانوباکتری، پیش و پس از بهینهسازی به روش اختلاط عصاره در محیط کشت بررسی شد. پس از بهینهسازی محیط کشت، درصد جلوگیری از رشد F. oxysporum f. sp. lycopersici از 33/79 به 43/87 درصد و برای AG2-2-IIIB R. solani از 47/82 به 62/90 درصد افزایش یافت. | ||
