| این پژوهش به بررسی استخراج پروتئین از دانههای خلر (Lathyrus sativus L.) و نیشی (Lathyrus rotundifolius L.) در شرایط مختلف pH و غلظتهای گوناگون نمک کلرید سدیم (NaCl) و همچنین در سیستمهای دو فازی آبی (ATPS) میپردازد. کمترین مقدار قابلیت استخراج پروتئین در pH برابر با ۴ و بیشترین آن در pH برابر با ۵/۹ با استفاده از آب مقطر بهدست آمد. نتایج نشان داد که افزودن نمک در شرایط قلیایی موجب افزایش معنادار قابلیت استخراج در مقایسه با محلولهای قلیایی بدون نمک نمیشود. در ادامه، سیستمی دوفازی بر پایه پلیاتیلن گلیکول (PEG) با وزن مولکولی ۴۰۰۰ گرم بر مول و صمغ عربی طراحی و برای استخراج و جداسازی پروتئینها از هر دو نوع دانه مورد ارزیابی قرار گرفت. در سیستم تشکیل یافته، PEG و صمغ عربی به ترتیب ترجیحاً به فازهای بالایی و پایینی مهاجرت کردند. بالاترین بازده استخراج، معادل با حدود ۶۳ درصد، در شرایط بهینه شامل ۲۰ درصد PEG، یک درصد صمغ عربی، نسبت جامد به مایع ۱:۱۰، اندازه ذرات با مش ۵۰ و دمای ℃ ۲۵ حاصل شد. پروفایلهای الکتروفورز (SDS-PAGE) الگوی تقسیمبندی وابسته به فاز واضحی را نشان داد که در آن زیرواحدهای با وزن مولکولی کم (۲۵ کیلو دالتون و کمتر) عمدتاً به فاز بالایی و زیرواحدهای با وزن مولکولی بالاتر (بیش از ۵۰ کیلو دالتون) به فاز پایینی مهاجرت کردند. نتایج این تحقیق نشان داد که سیستمهای ATPS روشی مؤثر، سازگار با محیطزیست و مقرونبهصرفه برای استخراج و تفکیک پروتئینها از دانههای خلر و نیشی هستند. | ||
