| Background :Testicular torsion is an urgent medical condition caused by the rotation of the spermatic cord, resulting in reduced blood flow to the testicle. Detorsion restores blood flow but may exacerbate testicular damage. Mesenchymal stem cells (MSCs), through their secretion of paracrine factors, ability to differentiate, and immunomodulatory effects, along with carvacrol, known for its anti-inflammatory and antioxidant benefits, may help alleviate infertility resulting from testicular torsion. Objective:This study investigates the effects of carvacrol and MSCs on testicular ischemia-reperfusion injury in rats. Methods:Fifty male Wistar rats were allocated into five different groups: Group 1 (no torsion), Group 2 (control, torsion-detorsion (TD) without treatment), Group 3 (TD + MSCs, weekly intraperitoneal MSC injections), Group 4 (TD + Carvacrol, daily intraperitoneal carvacrol injections for 21 days), and Group 5 (TD + Carvacrol + MSCs). Torsion involved a 720° rotation of the left testis under anesthesia for 3 hours. Following the final treatment, histopathological assessment was conducted using Johnson’s scoring system, along with oxidative stress tests Malondialdehyde (MDA (, Superoxide Dismutase )SOD (, Glutathione peroxidase (GPx), and Total antioxidant capacity (TAC). Statistical analysis was performed on the collected data. Results:The combination of carvacrol and MSCs notably decreased MDA levels and enhanced SOD and GPx levels when compared to the control group. (P < 0.05). These changes prevented testicular tissue damage and improved spermatogenesis in the injured testis. Conclusion:The synergistic effects of carvacrol and MSCs appear to effectively reduce oxidative stress and enhance antioxidant capacity, thereby mitigating ischemia-reperfusion-induced testicular damage. | ||
