بیان ژنهای سیتوکین در بافت طحال جوجههای گوشتی تغذیه شده با سطوح متفاوت آرد میوه بلوط | ||

چکیده | ||
جایگزینی ذرت با میوه بلوط در جیره طیور می تواند منجر به کاهش وابستگی و خروج ارز از کشور شود، اما بلوط دارای مقدار زیادی ترکیبات ضد تغذیه ای (تاننها) است که می تواند عامل محدود کنندهای در مصرف آن باشد. بنابراین بررسی اثرات مقادیر مختلف میوه بلوط در جیره بر عملکرد و سیستم ایمنی طیور ضروری می باشد تا سطوح مناسب بلوط برای استفاده در جیره طیور مشخص گردد. از این رو، هدف از این پژوهش، بررسی بیان ژنهای سیتوکین در بافت طحال جوجههای گوشتی تغذیه شده با جیره حاوی آرد میوه بلوط بود. به این منظور از سه جیره حاوی صفر، 15و 20 درصد آرد میوه بلوط برای تغذیه جوجههای گوشتی در یک دوره 42 روزه استفاده شد. در سنین 21 و 42 روزگی جوجهها، اندام طحال از تعداد 18 قطعه جوجه کشتار شده جدا گردید (از هر تیمار 6 عدد)، و از هر کدام RNA کل استخراج گردید و بیان ژنهای IL-2،IL-4 ، IL-5، IL-10، IL-13 وIFN- γ در هر 3 تیمار مشخص گردید. از ژن بتا اکتین نیز به عنوان ژن مرجع استفاده شد. دادههای حاصل از این بررسی با استفاده از نرم افزار REST 2009 V2.0.13 ارزیابی آماری گردید. با توجه به نتایج، بیان ژن IFN-γ در اندام طحال جوجههای گوشتی تغذیه شده با 15 درصد آرد میوه بلوط نسبت به گروه شاهد در هر دو سن 21 و 42 روزگی افزایش معنیداری داشت (p<0.05). در تیمار تغذیه شده با 20 درصد آرد میوه بلوط، اگرچه میزان بیان ژنIFN-γ در هر دو سن 21 و 42 روزگی، بالاتر از تیمار شاهد بود ولی این اختلاف معنیدار نبود. در سن 21 روزگی، بیان ژن IL-4 در جیره حاوی 15 درصد آرد میوه بلوط نیز افزایش معنیداری نسبت به گروه شاهد نشان داد (p<0.01). از طرف دیگر، در سن 42 روزگی، بیان ژنهای اینترلوکین 2 و 4 در تیمار 15 درصد آرد میوه بلوط نسبت به گروه شاهد کاهش معنیداری نشان داد (p<0.05). مقایسه بیان ژنهای IL-5، IL-10 و IL-13 در تیمارهای حاوی بلوط در هر دو سن 21 و 42 روزگی نسبت به گروه شاهد اختلاف معنیداری را نشان ندادند. بطور کلی، جایگزینی ذرت با بلوط در جیره جوجه های گوشتی، بر اساس مقدار و مدت زمان مورد استفاده، میتواند منجر به تغییر بیان ژنهای سیستم ایمنی در بافت طحال شوند. | ||
