غربال سودومونادهای فلورسنت بر پایة تولید سیدروفور و القای تولید اتیلن گیاه برای القای مقاومت سیستمیک علیه بیماری باکتریایی نواری برگ گندم | ||

چکیده | ||
در این تحقیق اثر سودومونادهای فلورسنت القاءکنندة مقاومت سیستمیک در کنترل بیماری باکتریایی نواری برگ گندم بررسی شد. با توجه به اهمیت سیدروفور (سیدروفور) تولیدشده توسط این باکتریها در القای مقاومت سیستمیک، محیط کشت CAS آگار (chrome azurol S) برای غربال باکتریهایی با بیشترین توان تولید سیدروفور استفاده شدند. از بین 200 جدایه که از گندمزار مختلف استانهای شمالغرب کشور جداسازی شدند، 37 جدایه توانایی بالایی در تولید سیدروفور داشتند و هالهای با قطر بیش از 15 میلیمتر روی محیط CAS ایجاد کردند. این جدایهها با آزمون تولید اتیلن بررسی شدند تا جدایههایی که قابلیت القای مقاومت سیستمیک روی گندم را دارند غربال شوند. از بین این جدایهها تنها چهار جدایه واکنشپذیری گیاه نسبت به Xanthomonas translucens pv. cerealis (XTC) را افزایش دادند و غلظت اتیلن تولیدشده در قطعههای برگی گیاهان تیمارشده با این جدایهها بیش از 2 پیکومول بر میلیلیتر هوا بود. تأثیر این جدایهها روی بیماری نواری برگ گندم در شرایط اتاقک کشت بررسی و مشخص شد که تنها جدایة 58A میتواند بهصورت معنیداری این بیماری را کاهش دهد. بررسی تغییرپذیری آنزیمی بوتههای تیمارشده با این جدایه در طول فرآیند بیماریزایی نشان داد که حضور این جدایه در ریشههای گندم تأثیری بر میزان فعالیت آنزیمهای پراکسیداز (PO) و فنیل آلانین آمونیالیاز (PAL) در بخش هوایی آن ندارد، اما پس از مایهزنی بیمارگر فعالیت هر دو آنزیم افزایش پیدا کرد. | ||
