| Background and purpose: A total of 2064 individuals of hematopoietic stem cell donors from various ethnic groups in Iran were included in this study. The primary objective of this research was to establish an HLA database and determine the genetic diversity among different ethnicities. Methods: DNA was extracted using the magnetic method. Subsequently, HLA-typing was conducted using the PCR-SSP method at a low-resolution level. Results were analyzed by independent experts, manually and via software. The HLA data from four different ethnicities, Gilak (n=510), Lor (n=465), Kurdish (n=719), and Arab (n=370), were examined for allele-ethnicity association. Allele frequencies were evaluated using statistical methods to determine any significant relationships. A significant level was set at 0.05. Additionally, standardized residuals were determined to identify the ethnicities in which the frequency of the studied alleles exceeded the expected values based on independence assumptions. Results: HLA-B*35 and HLA-B*51 exhibited the most prevalent allelic frequencies among HLA-B allele groups. It is worth mentioning that HLA-B*67, *78, *81, *82, and *83 were not observed in our study. Additionally, significant differences were detected in 17 alleles across the ethnic groups studied for HLA-B, with a P-value less than 0.05. Conclusion Determining the frequency of HLA alleles can specify similarities and differences in allelic groups within different ethnicities. This identification can be effective in designing an efficient strategy for establishing donor services in Iran's diverse regions and in stem cell registries. In the future, this data could lead to new clinical applications in transplantation, vaccine development, and infectious diseases. | ||
